Research Topic

Acute CNS injuries, including traumatic spinal cord injury and stroke, are devastating and dramatically life changing events with many potential complications and considerable mortality. While the incidence of stroke rapidly increases with age, traumatic spinal cord injury is disabling mostly young people. Despite intensive worldwide efforts, generally accepted clinical treatment for patients with CNS injury is not yet available. Up to date, much of the treatment of acute CNS injuries is only geared toward physical rehabilitation. Therefore, further intensive research is necessary and required also from economic and social aspects.

The initial (primary) insult of the spinal cord or brain initiates a cascade of destructive pathological mechanisms beginning immediately and lasting for weeks that spread from the epicenter to the surrounding tissue (secondary injury). These complex pathological processes represent a potential targets for various therapeutic interventions. Many of mechanisms are still unknown or explored insufficiently. The scientific research of our Department is therefore focused on better understanding of basic and pathological mechanisms, therapeutic control of pathological processes and stimulation of protective and regenerative endogenous mechanisms in order to reduce or prevent a neurological dysfunction resulting from these acute CNS injuries. The scientific research is organized and performed in three laboratories:

(1) The laboratory of neurochemistry and neurophysiology (Head: RNDr. N. Lukáčová, DrSc.)
(2) The laboratory of proteomics (Head: RNDr. P. Bonová, PhD.)
(3) The laboratory of electrophysiology and functional diagnostic (Head: RNDr. J. Gálik, CSc.).

Currently, our research is focused on the therapeutic control of secondary pathological mechanisms and stimulation of regenerative processes after traumatic spinal cord injury and cerebral ischemia by using the oscillating electrical field stimulation, endurance training, regulation of Angiotensin II receptor stimulation, the control of inflammatory response, the regulation of macrophages/microglia polarization, and the reduction of tissue damage after stroke by the remote conditioning (phenomena of ischemic tolerance). Due to the complexity of secondary damage and a limited ability of endogenous regenerative processes, our future strategy for effective therapy consists of appropriate combination of experimentally promising individual treatment approaches, and its translation to the clinical practice.
